(RDF) was used to treat the patients. The RDF was prepared by heating collagen in a microwave oven at a temperature of 1,000°C for 30 min. After cooling, the collagen was washed with water and dried in the sun for 1 h.
The collagen samples were then stored at −80° C until further analysis. For the analysis of collagen, collagenase was purified by using a modified method (Baker et al., 2000). The purified collagen is then subjected to a series of reactions to remove the free and bound collagen. These reactions include the addition of aqueous ammonium sulfate (ABS) to the reaction mixture, which is a mixture of ammoniacal and ammoniaacetic acids. Aqueously ammoniated collagen (AA) is the product of the AAB reaction. Amino acids are then added to this reaction to form the final product. This reaction is repeated several times to produce the total product, and the resulting product is purified. In the case of Rdf, a total of 10,500 μl of AAA was added into the RFF. To determine the concentration of free collagen and free AA, an aliquot of each sample was diluted with 1 ml of water. Then, 1 μL of this solution was mixed with 10 μM of total collagen A and AA. Finally, 10 μg of AA was dissolved in 10 ml water, washed, dried, then the solution containing the AA and collagen were added. All the samples (10 μm) were incubated at 37° to 40°c for 2 h, followed by centrifugation at 15,800 rpm for 10 min at 4° Celsius. Each sample (100 μml) of sample A was then incubation for 5 min in 0.1 M phosphate buffer (pH 7.4) and then washed. Samples B and C were washed and incubate for 15 min, respectively. Both samples of samples A, B, C and D were analyzed by HPLC. As shown in, the results showed that the amount of protein in samples B (0.5 μg/ml), C (1.0 μg) or D (2.2 μg), was significantly higher than that in sample D. However, no significant difference was found between the protein content of B or C. Therefore, it is concluded that Raffin’s Rapid collagen injection duplicates the proteins in collagen
