Oasis 7 Collagen Serum

oasis 7 collagen serum (PBS) and collagenase inhibitor (Ci) were added to the serum. The serum was then incubated for 30 min at room temperature with the following antibodies: anti-rabbit IgG (1:1000), anti–mouse IgA (2:100), and antihuman IgM (3:10). The samples were then washed twice with PBS and incubation was repeated with antirhodamine (0.1 mg/ml) for 1 h at 4°C. After washing, the samples was incubate for 2 h with a rabbit antimouse antibody (Rabbit-1; Sigma-Aldrich) at a concentration of 1:500.

The samples of the collagen-positive serum were analyzed by ELISA (Bio-Rad Laboratories, Hercules, CA) using the rabbit IgE-specific antibody, rabbit-2-phospho-Rhoda-Gly (r2P-gp) (Sigma- aldrick).
, and the total collagen content of serum samples from the patients were determined by the method of Kohn et al. (19). Serum samples (100 μl) of collagen were incubating with rabbit antiserum (10 μM) in PBS for 15 min. Then, samples containing collagen (50 μL) or collagenases (20 μg/mL) was added into the PBS. Serums were washed with phosphate-buffered saline (pH 7.4) twice and then the proteins were digested with 1% agarose gel (Invitrogen, Carlsbad, California) containing 1 μg of rabbit antibody. Protein digestion was carried out in a 96-well plate (Dako, San Diego, Calif.) with an ABI Prism 8500 (Applied Biosystems, Foster City, Ca.) and a 1.5% SDS-PAGE gels. A final concentration (5 μm) concentration was used for the analysis. For the determination of total protein content, serum from patients with collagen deficiency was diluted with 100 μmL of a solution of 0.2% Triton X-100 (Tris-Tec, St. Louis, Mo.) in phosphate buffered sodium phosphate buffer (PBST) supplemented with 10% fetal bovine serum albumin (FBS).
