zinc 100mg/kg, and the same dose of the other drugs was given to the rats. The rats were then killed by decapitation and decapitated with a sharp instrument. After decapitations, the brains were removed and stored at −80°C until further analysis.
The animals were divided into three groups: (i) the control group, (ii) rats treated with the drug and (iii) animals treated for the first time with either the drugs or saline. All animals in the group treated by the two drugs were killed immediately after the last injection. In the groups treated in (a) and in which the animals received the second drug, they were sacrificed at the end of 24 h. Animals in both groups were decapited and their brains removed. Brain sections were cut into 1.5-μm sections and stained with hematoxylin and eosin. Sections were stained for histone deacetylase (HDAC) activity using the anti-HDA antibody (Roche Diagnostics, St. Louis, MO). The sections of brains from the treated and control rats (n = 5) were stored in ice-cold liquid nitrogen at 4° C. for further histological analysis and for comparison with those of animals that received saline or the 2nd drug. Histological sections from animals receiving the saline treatment were also stained. For histology, sections (1.25-mm thick) of brain sections, from rats that were treated (control) or animals (2nd-drug) with saline were prepared and mounted on slides. Slides were mounted in a dark room and incubated at room temperature for 1 h at 37° K. and then transferred to a light-activated microscope (Nikon, Tokyo, Japan). Histology was performed using a Leica CX-100 microscope with an optical image stabilization system (Leica, Bologna, Italy).
