Cells were grown in Dulbecco’s modified Eagle’s medium (DMEM) supplemented with 10% fetal bovine serum (FBS) (Sigma- Aldrick) at 37°C. After 24 h, cells were washed twice with PBS and then incubated with primary antibodies (anti-CD3, anti-IL-1β, IL-6, TNFα, IFNγ, CD68, Bcl-2, p53, GAPDH, MCP-7, CXCL10, FGF21, caspase-9, NF-κB, nuclear factor-kappaB) for 24 hours at 4° C. Then, the cells and the medium were replaced with fresh medium. For the differentiation assay, a total of 24-h-old, nude mice were used. Mice were anesthetized with isoflurane and decapitated. A small amount of brain was removed and placed in ice-cold PBS. Following the decapitation, brain tissue was fixed with 2% paraformaldehyde in PBS for 1 hour. Brain was then removed, washed in 0.1 M PBS, incubation with anti–CD4 (0.5 μg/ml) or anti‐CD8 (10 μg) was performed for 30 minutes at room temperature. Next, 1 ml of 1% Triton X-100 (Pierce)
